Development of 3-D chromatin texture analysis

چکیده
The aim of the study described in this Chapter was to develop and implement 3-D texture features that quantitatively describe the nuclear chromatin distribution. Thirty-five features thoughtfully chosen from 4 categories of 3-D texture features (discrete texture features, Markovian features, fractal features, grey value distribution features) were selected and tested for invariance properties (rotation and scaling) using artificial images with a known grey value distribution. The discriminative power of the 3-D texture features was tested on artificially constructed benign and malignant 3-D nuclei with increasing nucleolar size and advancing chromatin margination towards the periphery of the nucleus. 10 Benign and 10 malignant human prostate nuclei were acquired by CLSM to evaluate whether there was more texture information present in 3-D whole nuclei compared to a single 2-D plane from the middle of the nucleus. A set of 35 3-D nuclear texture features was used successfully to assess nuclear chromatin patterns in 3-D images obtained by confocal laser scanning microscopy. Racial Differences in 3-D Nuclear Chromatin Patterns of Prostate Cancer
Purpose: There is a significant difference in prostate cancer incidence and stage corrected mortality between African-American (AA) and Caucasian-American (CA) men. These differences have largely been contributed to socialeconomic factors, yet variation in prostate cancer related gene expression has been found as well. The aim of this study was to analyze whether these differences are reflected...
متن کاملSperm nuclear texture assessment: a fertility prediction tool Introduction Sperm nuclear morphometry quantifies properties of the nucleus and allows comparison between semen
Sperm nuclear morphometry quantifies properties of the nucleus and allows comparison between semen samples. Chromatin packaging is one of the properties reflected by nuclear measures, and has been related to fertility as correct packaging is essential for not only fertilization but also successful early embryonic development. One such morphometric measure is texture. Nuclear texture is thought ...
متن کاملDevelopment of 3D Chromatin Texture Analysis Using Confocal Laser Scanning Microscopy
INTRODUCTION Analysis of nuclear texture features as a measure of nuclear chromatin changes has been proven to be useful when measured on thin (5-6 microm) tissue sections using conventional 2D bright field microscopy. The drawback of this approach is that most nuclei are not intact because of those thin sections. Confocal laser scanning microscopy (CLSM) allows measurements of texture in 3D re...
متن کاملEtiology and Evaluation of Sperm Chromatin Anomalies
Evidence suggests that human sperm chromatin anomalies adversely affect reproductive outcomes and infertile men possess substantially amount of sperm with chromatin anomalies than fertile men. Routine semen analysis evaluates parameters such as sperm motility and morphology, but does not examine the nuclear DNA integrity of spermatozoa. It has been suggested that altered nuclear chromatin struc...
متن کاملMolecular characteristics and chromatin texture features in acute promyelocytic leukemia
BACKGROUND Acute promyelocytic leukemia is a cytogenetically well defined entity. Nevertheless, some features observed at diagnosis are related to a worse outcome of the patients. METHODS In a prospective study, we analyzed peripheral (PB) leukocyte count, immunophenotype, methylation status of CDKN2B, CDKN2A and TP73; FLT3 and NPM1 mutations besides nuclear chromatin texture characteristics ...
